No, you do not have to fly on an online network, but (at the moment) you do have to maintain an internet connection for the duration of the flight.
To lock a flight, go to Divisions/ProPilot. Log in, and click Lock Flight on the menu at the top. Find a plane at the airport where you wish to start, and click on its Tail ID to select a flight. Happy flying
